Quantification of cells or microbes by quantitative (qPCR) and of gene expression by reverse-transcriptase qPCR (RT-qPCR) is common practice in molecular labs. However, qPCR has several limitations, including the need for an external standard curve for absolute quantification, hampering direct comparisons between different studies, and pronounced technical variation at low template densities. Droplet digital PCR (ddPCR) offers an alternative method for quantification without the need for a standard curve and greatly reducing technical variability. ddPCR has become a gold standard in nucleic acid quantification, studies on copy number variation, and other applications.
The Koepfli lab uses ddPCR routinely for a range of applications. This seminar will provide an introduction into the principle of ddPCR and discuss several applications. The aim of this seminar is to equip all participants with what is needed to run a ddPCR experiment successfully.
