Seminar Title: "A large genome-wide association study of prostate specific antigen"
Abstract: In this research + teaching talk, I will introduce how genome wide association studies (GWAS) are conducted and highlight what researchers can learn from GWAS. Then I will give an example from my recent work on a GWAS meta-analysis of prostate-specific antigen (PSA). PSA screening for prostate cancer has been controversial due to over-treatment. Prior efforts have suggested that screening may be improved with genetic adjustment, but have been focused primarily on largely European ancestry populations, and many loci remain to be discovered. We conducted a multi-ancestry genome-wide meta-analysis of nearly 300K men, identifying ~300 genome-wide significant variants, ~200 of which are novel, most with some evidence of replication in a separate cohort. These results increase the variance explained of genome-wide significant variants, up to a total of 6-12% depending on the population ancestry, improving the future potential for adjusting PSA testing to reduce unnecessary biopsies.
